Powder and excipients: what the appearance of a vial reveals
A broken puck, more powder, or a clear solution do not measure purity or quantity. Learn to separate appearance, gross mass and content.

The freeze-dried material can be viewed as a layer, a compact structure, or a powder. GenScript explains that freeze-drying conditions and composition influence the appearance, without the visual difference necessarily implying a difference in mass.
This does not normalize any observation: unexpected humidity, foreign particles or a damaged seal must be recorded as an incident. Ask for the appearance criteria of the reference and document the batch. A photograph can support the query, but it alone cannot resolve its cause.

Gross mass, net content and added materials
The weight of a preparation may include moisture, counterions, or other specified materials. Bachem distinguishes raw mass and peptide content; that difference explains why an HPLC area percentage does not automatically convert the entire weight into pure peptide.
If an excipient or formulation agent is declared, request its identity and quantity basis. Do not assume that it is present because you see more dust, nor that it is absent because you see less. You need information about the specific preparation and a clear definition of what the quantity on the label means.
Transparency, turbidity and color
A transparent solution may lack identity, content or sterility data. A cloudy solution may require solubility investigation or other issues. Neither observation offers a complete diagnosis.
Records reference, status, date, conditions and what changed. Do not take steps to modify material based on a photo or anonymous response. Contrast with the record and the person responsible for the essay.
Reception record: what to keep
Photograph label, closure and packaging when relevant, without losing the lot. Note whether the product was received dry or in solution and the conditions indicated by the supplier. Also keep delivery note and documentation; The observed state must be able to be related to a delivery.
If the project needs quantification, ask for a content method. Comparing the volume of powder from two photographs or weighing an entire vial with its container does not answer what mass of compound is inside.

Does weighing the entire vial verify the mg of peptide?
No. It includes the container and does not separate the components of the preparation. Request an essay of appropriate content.
Does the absence of visible changes prove stability?
No. Stability must be related to the reference, conditions and available data or tests.
